A relationship between antimicrobial peptide gene expression and capacity of a selected shrimp line to survive a Vibrio infection ArchiMer
De Lorgeril, Julien; Gueguen, Yannick; Goarant, Cyrille; Goyard, Emmanuel; Mugnier, Chantal; Fievet, Julie; Piquemal, D; Bachere, Evelyne.
Understanding of antimicrobial defence mechanisms of penaeid shrimp should help in the design of efficient strategies for the management and disease control in aquaculture. In this study, we have specifically analysed the expression in circulating hemocytes of antimicrobial peptides (AMPs) encoding genes, such as PEN2 and PEN3, ALF, crustin, lysozyme and a putative cysteine-rich peptide. We evidenced a relationship between the level of expression of some AMPs and the successful response of the shrimp, Litopenaeus stylirostris, to circumvent a pathogenic Vibrio penaeicida infection. Additionally, significant differences in some AMP transcript amounts are evidenced between control, non-selected shrimp line and the third generation breeding of shrimp selected...

Cloning, purification and comparative characterization of two digestive lysozymes from Musca domestica larvae BJMBR
Cançado,F.C.; Chimoy Effio,P; Terra,W.R.; Marana,S.R..
cDNA coding for two digestive lysozymes (MdL1 and MdL2) of the Musca domestica housefly was cloned and sequenced. MdL2 is a novel minor lysozyme, whereas MdL1 is the major lysozyme thus far purified from M. domestica midgut. MdL1 and MdL2 were expressed as recombinant proteins in Pichia pastoris, purified and characterized. The lytic activities of MdL1 and MdL2 upon Micrococcus lysodeikticus have an acidic pH optimum (4.8) at low ionic strength (μ = 0.02), which shifts towards an even more acidic value, pH 3.8, at a high ionic strength (μ = 0.2). However, the pH optimum of their activities upon 4-methylumbelliferyl N-acetylchitotrioside (4.9) is not affected by ionic strength. These results suggest that the acidic pH optimum is an intrinsic property of...

Effect of fish meal replacement by plant protein sources on non-specific defence mechanisms and oxidative stress in gilthead sea bream (Sparus aurata) ArchiMer
Sitja Bobadilla, A; Pena Llopis, S; Gomez Requeni, P; Medale, Francoise; Kaushik, Sadasivam; Perez Sanchez, J.
Partial or total replacement of fish meal by a mixture of plant protein (PP) sources (corn gluten, wheat gluten, extruded peas, rapeseed meal and sweet white lupin) balanced with indispensable amino acids was examined in juvenile gilthead sea bream over the course of a 6-month growth trial. A diet with fish meal (FM) as the sole protein source was compared to diets with 50%, 75% and 100% of replacement (PP50, PP75, and PP100). The possible influence of diets on growth performance, plasma metabolites, gut integrity, liver structure, anti-oxidant and immune status was evaluated. Final body weight was progressively decreased with PP inclusion, but in PP50 and PP75-fed fish, feed efficiency (FE) was significantly improved and specific growth rates remained...

Effect of lysozyme or nisin on survival of some bacteria treated with high pressure at subzero temperature BJM
Malinowska-Pańczyk,Edyta; Kołodziejska,Ilona.
The aim of this work was to examine the inactivation of some Gram-positive and Gram-negative bacteria exposed to the pressure of 193 MPa at -20 °C in the presence of lysozyme or nisin at concentration of 400 mg/ml. The highest effect of pressure at subzero temperature and lysozyme was found with pressure sensitive Pseudomonas fluorescens; viable cells of this strain were not detected in 1 ml of sample after combined treatment. The action of pressure at subzero temperature and lysozyme or nisin against Escherichia coli led to synergistic reduction by 0.7 or 1.6 log cycles, respectively, while it was practically insignificant for two Staphylococcus aureus strains. Viability loss of E. coli and S. aureus occurred during storage for 20 h of the samples at 37...

Etude de la variabilité spatio-temporelle des caractéristiques physiologiques des jeunes stades de vie de la seiche Sepia officinalis L. en Manche. ArchiMer
Safi, Georges.
The common cuttlefish Sepia officinalis, is a major exploited marine resource in the English Channel. It has strong seasonal and interannual variations in recruitment. Mechanisms affecting the renewal of the stock by the arrival of new “recruits” and their number need to be better understood. Thus the study of the spatial and temporal variability of “pre-recruits” physiological characteristics was conducted in four spawning sites of the Channel [i.e. Agon Coutainville (FR), Baie de Seine (FR), Selsey (UK) and Torbay (UK)]. The studied parameters were the eggs quality and hatching rate then, the digestive and immune performance of juveniles in relation to local environmental conditions. English eggs have a delayed hatching time and higher hatching rate when...

Immunomodulation of Juvenile Pacu, Piaractus mesopotamicus, by Different β(1-3)(1-6)-D glucan Products BABT
Sabioni,Rafael Estevan; Zanuzzo,Fábio Sabbadin; Cyrino,José Eurico Possebon.
Abstract Stress in intensive fish farming hamper immune function of fish and cause losses by disease outbreaks, a situation that can be minimized, but cannot be completely circumvented, by the use of immunomodulators. Addition of immunomodulators to aquafeeds has thus become a common practice. β-glucan (BG) is one of most studied and effective immunomodulators, aquaculture purposes included. Extracted from cell walls of bacteria, fungi and selected cereals, BG activity depends on the source and extraction methods. This study evaluated effects of two BG products (BG1 and BG2), extracted from Saccharomyces cerevisiae under varying extraction methods and with different immune activity, on the feeding of pacu Piaractus mesopotamicus juveniles. BG1 provided...

Immunostimulation and increase of intestinal lactic acid bacteria with dietary mannan-oligosaccharide in Nile tilapia juveniles R. Bras. Zootec.
Levy-Pereira,Nycolas; Yasui,George Shigueki; Cardozo,Marita Vedovelli; Dias Neto,José; Farias,Thaís Heloísa Vaz; Sakabe,Roberson; Pádua,Santiago Benites de; Pilarski,Fabiana.
ABSTRACT In this experiment, the beneficial effects of mannan-oligosaccharide (MOS) on immunology and intestinal microbiology of Nile tilapia juveniles was demonstrated. Prior to this, three levels of MOS in Nile tilapia diets (1, 8, and 15 g.kg−1) were tested, and hematological parameters, serum lysozyme, and intestinal microbiology were analyzed. The fish blood was sampled at day zero (basal sample) and after 45 days of trial, and the intestinal microbiota was evaluated at the end of the experiment. After 45 days of trial, fish fed 8 and 15 g.kg−1 of MOS presented an increase in both aerobic and lactic acid bacteria numbers in their guts. The MOS feeding also increased the counts of total leukocytes, monocytes, and lymphocytes of fish, but a decrease in...

Influence of temperature, salinity and E. coli tissue content on immune gene expression in mussel: Results from a 2005-2008 survey ArchiMer
Li, Hui; Toubiana, Mylene; Monfort, Patrick; Roch, Philippe.
Several bivalves, including mussels, suffered from mortalities particularly in summer. To look for the possible effect of environmental parameters on immune capacities, Mytilus galloprovincialis were collected monthly from August 2005 to July 2008 from the Palavas Laguna, French Mediterranean coast. Q-PCR was used to quantify the expression of three antimicrobial peptide genes (defensin, mytilin B and myticin B), in addition to lysozyme and HSP70. House keeping gene was 28S rRNA. Defensin, myticin B and lysozyme appeared more expressed in spring-summer than in winter. In contrast, HSP70 expression was higher in winter. Statistical studies using principal component analysis (PCA) and multiple regression models revealed positive influence of temperature on...

Lysozyme gene expression and hemocyte behaviour in the Mediterranean mussel, Mytilus galloprovincialis, after injection of various bacteria or temperature stresses ArchiMer
Li, H; Parisi, M; Toubiana, Mylene; Cammarata, M; Roch, Philippe.
The aim of the present study was to evaluate the expression of the Mytilus galloprovincialis lysozyme gene in different in vivo stress situations, including injection of bacteria Vibrio splendidus LGP32, Vibrio anguillarum or Micrococcus lysodeikticus, as well as heat shock at 30 C and cold stress at 5 C. Injection of V. splendidus LGP32 resulted in: (i) a general down-regulation of lysozyme gene expression, as quantified by Q-PCR; (ii) reduction in the number of circulating hemocytes; (iii) decrease in the percentage of circulating hemocytes expressing lysozyme mRNA which was now restricted to only small cells, as observed by ISH; and (iv) accumulation of hemocytes expressing lysozyme in the muscle sinus where injection took place. Injection of V....

Practical use of immobilized lysozyme for the remediation process of Escherichia coli in aqueous solution Electron. J. Biotechnol.
Alves,Carlucio R; Pimenta,Maria Gardenny R; Vieira,Regine H.S.F; Furtado,Roselayne F; Guedes,Maria Izabel F; Silva,Rui C.B; Assis,Odilio B.G.
The lysozyme enzyme was immobilized on vitreous surface (fragments with diameters of 0.3 and 1.0 mm) for remediation of the microorganism Escherichia coli JM 109 into fresh water and saline solutions with 0.9% NaCl (w/v). Characterization of enzymatic film was carried out by infrared spectroscopy and atomic force microscopy techniques. Bactericide activity of the enzyme was evaluated by spectrophotometric analysis. It was verified that the enzymatic film was strongly coupled with the vitreous surface. The topographic analysis demonstrated that the deposited film was uniform and homogeneous. It was observed bactericide activity of film deposited on vitreous surface with 0.3 mm in fresh and saline solutions. This fact was not verified to vitreous fragments...

Pressure-assisted cold denaturation of hen egg white lysozyme: the influence of co-solvents probed by hydrogen exchange nuclear magnetic resonance BJMBR
Vogtt,K.; Winter,R..
COSY proton nuclear magnetic resonance was used to measure the exchange rates of amide protons of hen egg white lysozyme (HEWL) in the pressure-assisted cold-denatured state and in the heat-denatured state. After dissolving lysozyme in deuterium oxide buffer, labile protons exchange for deuterons in such a way that exposed protons are substituted rapidly, whereas "protected" protons within structured parts of the protein are substituted slowly. The exchange rates k obs were determined for HEWL under heat treatment (80ºC) and under high pressure conditions at low temperature (3.75 kbar, -13ºC). Moreover, the influence of co-solvents (sorbitol, urea) on the exchange rate was examined under pressure-assisted cold denaturation conditions, and the corresponding...

Recombinant expression and refolding of the c-type lysozyme from Spodoptera litura in E. coli Electron. J. Biotechnol.
Kim,Jong-Wan; Yoe,Jeehyun; Lee,Gil Ho; Yoe,Sung Moon.
The chicken-type lysozyme of the insect Spodoptera litura (SLLyz) is a polypeptide of 121 amino acids containing four disulfide bridges and 17 rare codons and participates in innate defense as an anti-bacterial enzyme. The recombinant S. litura lysozyme (rSLLyz) expressed as a C-terminal fusion protein with glutathione S-transferase (GST) in Rosetta(DE3) Singles. The protein was produced as an inclusion body which was solubilized in 8 M urea, renatured by on-column refolding, and purified by reversed-phase chromatography to 95% purity. The purified rSLLyz demonstrated antibacterial activity against B. megaterium confirmed by inhibition zone assay. The overexpression and refolding strategy described in this study will provide a reliable technique for...

The chromatin structure of the lysozyme GAS41 origin of DNA replication changes during the cell cycle Biol. Res.
ZIMMERMANN,KATRIN; HOLTZ,MARLIS; PHI-VAN,LOC.
We used a rapid and simple protocol using lysolecithin for mapping HS sites in vivo. The protocol is based on partial digestion with DNase I of exponentially growing cells following permeabilization by short treatment with lysolecithin. Using this protocol, we analyzed the chromatin structure of the region surrounding two overlapping elements, an origin of bidirectional DNA replication and the GAS41 promoter, in chicken myelomonocytic HD11 cells arrested in G0, G0 and S phases as well as at the G0/S border. The results show that the chromatin of this region became more nuclease sensitive when cells were arrested in G0 phase and that this change in chromatin structure was reversible after the cells began to enter S phase
